You have to change/configure the MIDI Port you have selected at Tools -> Settings, at sound tab.
That default MIDI Port, as different ports you could configure, are OS dependant.. so, what is your operating system ?

If you have a windows release, you should have the fluidsynth plugin installed
Check it at menu "Tools -> Plugins", select "Fluidsynth output plugin", and click configure.
at the opened window you may find options to add soundfonts ( *.sf2 files )
You have to download one GM (general midi) standard soundfont,
here there are 2 you can test: ftp://ftp.personalcopy.net/pub/Unison.sf2.gz
ftp://ftp.personalcopy.net/pub/PC51f.sf2.gz
make sure to uncompress them, and add one of them (or both ) to the fluidsynth plugin..
once you have your soundfont configured, you can go again to "Tools -> Preferences", and at MIDI Port drop down, you may see a new option available such as "TG Fluidsynth [soundfont_name]"

hmm if fluidsynth plugin gives you problems, you could try installing gervill.
Gervill is a software synthesizer implementation for java.
Download it from:
https://gervill.dev.java.net/
To install it, you have to find where java is installed ( often inside Program Files/ ) where java is installed, you have to find a folder structure like "jre/lib/ext/" , so you have to paste the gervill.jar file inside that "ext" folder.
Then next time you start tuxguitar, at midi ports you have to see the "Gervill" option.
then next step is configure the soundfont to it.. you have to go again to "Tools -> Plugins" but in this case, select "Java sound api plugin" and click configure.
make sure you check "Use custom soundbank", and then select the .sf2 file.

Try this one:
https://gervill.dev.java.net/examples/gervill.jar
This file must be inside lib/ext of your JRE folder.
